Materials and methods

 

This project was achieved on 3 commercial varieties of tobacco which are widely grown throughout the world:  K326 for the Flue-cured type, Burley 21 and TN86 for the Burley type.  In order to maximise the number of different unique sequences, mRNA from various organs of the plant (seeds, roots, stem, midrib, lamina and flowers) was isolated at different stages of development (germination, young seedlings, before and after topping, maturity). 

11 cDNA libraries were created and 56 000 clones were obtained during this project.

11/20/06: the bacterial clones are available at the CNRGV Toulouse.


07/12/06: a unigene set and annotations are available on our website.


04/27/06: the sequencing of 56000 clones is completed. 46546 EST sequences are freely available to the public in EMBL and Genbank databases.


12/09/05: 6515 additional EST sequences increase the total number of available sequences to 11776.


10/03/05: 5261 new EST sequences available in EMBL and Genbank databases.
